Immune Checkpoint Inhibitors in EGFR-Mutated NSCLC: Dusk or Dawn?

نویسندگان

چکیده

Although immune checkpoint inhibitors (ICIs) that target programmed cell death protein-1/programmed ligand-1 axis have significantly shifted the treatment paradigm in advanced NSCLC, clinical benefits of these agents are limited patients with EGFR-mutated NSCLC. Several predictive biomarkers (e.g., expression, tumor mutation burden), which been validated EGFR-wild type however, not efficacious tumors, suggesting unique characteristics microenvironment Here, we first summarized evidence on efficacy ICIs Then, cancer immunogram features NSCLC was depicted to visualize state cancer-immune system interactions, including foreignness, sensitivity effectors, metabolism, general status, infiltration, cytokines, and soluble molecules. We further discussed potential subpopulations EGFR mutations could benefit from ICI treatment. Lastly, put forward future strategies adequately maximize upcoming era combination immunotherapies.

Gefitinib Treatment in EGFR Mutated Caucasian NSCLC

INTRODUCTION In the phase IV, open-label, single-arm study NCT01203917, first-line gefitinib 250 mg/d was effective and well tolerated in Caucasian patients with epidermal growth factor receptor (EGFR) mutation-positive non-small-cell lung cancer (previously published). Here, we report EGFR mutation analyses of plasma-derived, circulating-free tumor DNA. METHODS Mandatory tumor and duplicate ...

Outcome of EGFR-mutated NSCLC patients with MET-driven resistance to EGFR tyrosine kinase inhibitors

Background Several mechanisms of acquired resistance to EGFR tyrosine kinase inhibitors (TKIs) in EGFR-mutated NSCLC have been described including the T790M mutation and MET amplification. Whereas T790M mutation confers prolonged survival and sensitivity to 3rd generation TKIs, data are lacking on clinical features and outcome of MET-driven resistant EGFR-mutated NSCLC patients. Methods Patie...

Beyond disease-progression: Clinical outcomes after EGFR-TKIs in a cohort of EGFR mutated NSCLC patients

PURPOSE Treatment and clinical-outcomes were described in a sub-cohort of non-small-cell lung cancer (NSCLC) patients with disease-progression (PD) after epidermal growth factor tyrosine kinase inhibitors (EGFR-TKIs) treatment. PATIENTS AND METHODS We retrospectively analyzed a single-institutional EGFR mutation positive (EGFRmut+) NSCLC cohort for post-TKI-PD management, and assessed overall...

Cardiotoxicity of immune checkpoint inhibitors

Cardiac toxicity after conventional antineoplastic drugs (eg, anthracyclines) has historically been a relevant issue. In addition, targeted therapies and biological molecules can also induce cardiotoxicity. Immune checkpoint inhibitors are a novel class of anticancer drugs, distinct from targeted or tumour type-specific therapies. Cancer immunotherapy with immune checkpoint blockers (ie, monocl...

Immune checkpoint inhibitors in cancer therapy.

In recent years immune checkpoint inhibitors have garnered attention as being one of the most promising types of immunotherapy on the horizon. There has been particular focus on the immune checkpoint molecules, cytotoxic T-lymphocyte antigen-4 (CTLA-4) and programmed cell death protein 1 (PD-1) which have been shown to have potent immunomodulatory effects through their function as negative regu...
